CFTR (HGNC:1884): (CF transmembrane conductance regulator) This gene encodes a member of the ATP-binding cassette (ABC) transporter superfamily. The encoded protein functions as a chloride channel, making it unique among members of this protein family, and controls ion and water secretion and absorption in epithelial tissues. Channel activation is mediated by cycles of regulatory domain phosphorylation, ATP-binding by the nucleotide-binding domains, and ATP hydrolysis. Mutations in this gene cause cystic fibrosis, the most common lethal genetic disorder in populations of Northern European descent. The most frequently occurring mutation in cystic fibrosis, DeltaF508, results in impaired folding and trafficking of the encoded protein. The variant allele was found at a frequency of 4e-06 in 251332 control chromosomes. c.1466C>A has been reported in the literature in a homozygous individual and multiple compound heterozygous individuals affected with Cystic Fibrosis (Example: DeBie_2012). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 22627569). Seven clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This changes the amino acid from a serine to a stop codon within coding exon 11. In one study, this mutation was described as compound heterozygous in 12 individuals with another pathogenic mutation and as homozygous in one individual all with classic cystic fibrosis (De Bie I et al. Genet Med. 2012;14(10):883-886). This mutation has also been observed in an individual with congenital bilateral absence of the vas deferens (Steiner B et al. Hum Mutat. 2011;32(8):912-20). Affected individuals carrying this pathogenic mutation had elevated sweat chloride levels, pancreatic insufficiency, and higher rates of Pseduomonas infection (Sosnay PR et al. Nat Genet. 2013;45(10):1160-1167 and The Clinical and Functional Translation of CFTR (CFTR2); available at http://cftr2.org. Accessed June 13, 2022). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
