Variant summary: CFTR c.1811C>G (p.Thr604Ser) results in a conservative amino acid change located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. Several computational tools predict a significant impact on normal splicing: Three predict the variant creates a 5' donor site. A similar splicing change was reported in the literature in a patient within a CFTR database (Ramalho_2016). The variant was absent in 236058 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1811C>G has been observed in a compound heterozygous individual affected with Cystic Fibrosis also carrying the pathogenic F508del variant (Schrijver_2005). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 25735457, 34996830, 15858154). ClinVar contains an entry for this variant (Variation ID: 53394).
