Variant summary: CFTR c.2215delG (p.Val739TyrfsX16) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant allele was found at a frequency of 4e-06 in 251182 control chromosomes. c.2215delG has been well reported in the literature in multiple individuals affected with Cystic Fibrosis (example, Sosnay_2013). These data indicate that the variant is very likely to be associated with disease. No experimental evidence demonstrating an impact on protein function was ascertained. Two clinical diagnostic laboratories and one expert panel (CFTR2) have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All submitters classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
ClinVar contains an entry for this variant (Variation ID: 53455). This variant is also known as c.2347delG. For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Val739Tyrfs*16) in the CFTR gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CFTR are known to be pathogenic (PMID: 1695717, 7691345, 9725922). This variant is present in population databases (rs397508353, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with cystic fibrosis and pancreatic insufficiency (PMID: 10439967, 22658665, 23974870). -
The c.2215delG pathogenic mutation, located in coding exon 14 of the CFTR gene, results from a deletion of one nucleotide at nucleotide position 2215, causing a translational frameshift with a predicted alternate stop codon (p.V739Yfs*16). This mutation was identified in one individual with cystic fibrosis; however, complete genotype information was not provided (Soltysova A et al. Clin Respir J, 2017 May;doi: 10.1111/crj.12651. [Epub ahead of print]). This mutation is associated with pancreatic insufficiency and elevated sweat chloride levels (Sosnay PR et al. Nat. Genet., 2013 Oct;45:1160-7). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. This amino acid position is well conserved in available vertebrate species. -
