Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251070 control chromosomes (gnomAD). c.3971T>C has been reported in the literature and in CF databases as a biallelic genotype in individuals affected with Cystic Fibrosis (e.g. Bienvenu_2005, McCague_2019, SickKids, CFTR-France). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 16596947, 20163773, 30888834). Two submitters, including the expert panel CFTR2, have cited clinical-significance assessments for this variant to ClinVar after 2014.
