Variant summary: DSP c.7623delG (p.Lys2542SerfsX19) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251490 control chromosomes (gnomAD). c.7623delG has been reported in the literature in multiple homozygous individuals affected with Carvajal Syndrome (Norgett_2000). These data indicate that the variant is very likely to be associated with disease. Two ClinVar submitters (evaluation after 2014) cite the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

ClinVar contains an entry for this variant (Variation ID: 16837). This variant is also known as 7901delG. This premature translational stop signal has been observed in individuals with Carvajal syndrome (PMID: 11063735). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Lys2542Serfs*19) in the DSP gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 330 amino acid(s) of the DSP protein. -
