This missense variant replaces aspartic acid with asparagine at codon 151 of the MYBPC3 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been performed for this variant. This variant has been observed in several individuals affected with hypertrophic cardiomyopathy (PMID: 27532257, 28356264, 28771489, 33495597) or dilated cardiomyopathy (PMID: 32746448). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 151 of the MYBPC3 protein (p.Asp151Asn). The Asp151Asn variant has not been reported in the literature. In addition, this variant is not well-conserved in mammals, suggesting that the change may be wel l tolerated. Of note, this individual's race is reported to be Black and our lab oratory has only tested around 130 Black probands to date. Therefore, we have li mited data on variants found in the Black population. Finally, this variant has never been seen in isolation in a proband. As such, the clinical significance of this variant cannot be determined at this time. -
