The 506-1G>T variant in MYBPC3 has been identified by our laboratory in 1 indivi dual with HCM (LMM unpublished data). This variant occurs in the invariant regio n (+/- 1,2) of the splice consensus sequence and is predicted to cause altered s plicing leading to an abnormal or absent protein. Truncating variants in MYBPC3 are established as pathogenic for HCM. In summary, this variant meets our criter ia for pathogenicity (http://pcpgm.partners.org/LMM). -
