The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism, and it is reported as pathogenic/likely pathogenic by multiple laboratories in ClinVar (Variation ID: 43595). This variant abolishes the canonical splice acceptor site of intron 8, which is likely to disrupt gene function. Other variants that abolish the same splice acceptor site (c.920-1G>A, c.920-1G>T) have been reported in individuals with symptoms or diagnoses of Li-Fraumeni syndrome, suggesting that disruption of this splice site is associated with disease (Gonzalez 2009, Wu 2011).
