The leucine at codon 908 is replaced by proline, an amino acid with similar properties. This alteration has been reported in association with dilated cardiomyopathy (DCM) (Parikh VN et al. Circ Heart Fail, 2019 Mar;12:e005371; Horvat C et al. Genet Med, 2019 Jan;21:133-143). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
