This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of dilated cardiomyopathy (PMID: 29892087). ClinVar contains an entry for this variant (Variation ID: 43994). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on RBM20 protein function. The p.L908P variant (also known as c.2723T>C), located in coding exon 11 of the RBM20 gene, results from a T to C substitution at nucleotide position 2723. The leucine at codon 908 is replaced by proline, an amino acid with similar properties. This alteration has been reported in association with dilated cardiomyopathy (DCM) (Parikh VN et al. Circ Heart Fail, 2019 Mar;12:e005371; Horvat C et al. Genet Med, 2019 Jan;21:133-143). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
