The Gly453Ala variant (ABCC9) has not been reported in the literature but has be en detected by our laboratory in 1 individual with DCM (this individual carried another, likely pathogenic DCM variant). Glycine (Gly) at position 453 is highly conserved in evolution, increasing the likelihood that change would not be tole rated. Computational tools (AlignGVGD, PolyPhen2, SIFT) also predict that a chan ge to alanine (Ala) would impact the protein, though their accuracy is unknown. Additional information is needed to fully assess the clinical significance of th e Gly453Ala variant. -
