Variant summary: TTN c.86925A>G (p.Ile28975Met) results in a conservative amino acid change located in the A band region of the encoded protein sequence. Two of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 7.7e-05 in 247874 control chromosomes in the gnomAD database, including 1 homozygotes suggesting a benign role for this variant. c.86925A>G has been reported in the literature in individuals affected with cardio myopathy and epilepsy (example: Pugh_2014 and Allen_2021). At-least one of these reports classified the variant as VUS (Pugh_2014). These report(s) do not provide unequivocal conclusions about association of the variant with Limb-Girdle Muscular Dystrophy, Type 2J. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
