The BRCA2 c.7651A>C (p.Lys2551Gln) variant has been reported in the published literature in an unaffected individual (PMID: 32467295 (2020), see also LOVD (https://databases.lovd.nl/shared/variants/BRCA2)). In addition, in silico analyses have described the variant with conflicting classifications (PMID: 25348012 (2014), 30212499 (2018)). The frequency of this variant in the general population, 0.000008 (2/251118 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

This variant has been reported in a carrier family in an ENIGMA multifactorial analysis but a final classification and clinical history was not described (PMID: 31131967). This variant has been identified in 2/251118 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
