The encoded protein is part of a large protein complex called ASCOM, which has been shown to be a transcriptional regulator of the beta-globin and estrogen receptor genes. The observed frameshift variant c.4135_4136del(p.Met1379ValfsTer52) in the KMT2D gene has been reported previously in individuals with Kabuki syndrome (Cheon CK, Ko JM., 2015; Bögershausen N, et al., 2016). This variant is absent in the gnomAD Exomes. This variant causes a frameshift starting with codon Methionine 1379, changes this amino acid to Valine residue, and creates a premature Stop codon at position 52 of the new reading frame, denoted p.Met1379ValfsTer52. It is submitted to ClinVar as Pathogenic/Likely pathogenic. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing. For these reasons, this variant has been classified as Pathogenic. -
