The isoleucine (Ile) residue at position 4996 is poorly conserved across most species, including mammals, and computational analy ses (biochemical amino acid properties, AlignGVGD, PolyPhen2, and SIFT) suggest that the Ile4996Phe variant may not impact the protein. However, this informatio n is not predictive enough to rule out pathogenicity. In summary, the clinical s ignificance of this variant cannot be determined with certainty; however based u pon the conservation and computational data, we would lean towards a more likely benign role. -
