The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.371C>T has been reported in the literature in at least one individual affected with personal or family history of breast cancer (e.g. Rahman_2007). This report does not provide unequivocal conclusions about association of the variant with Breast Cancer. At least one publication reports experimental evidence evaluating an impact on protein function in a mammalian two-hybrid assay showing mildly reduced (75% of WT) BRCA1 binding activity in vitro (e.g. Rodrigue_2019).
