The variant of interest was observed in the large, broad control population, ExAC, with an allele frequency of 4/120564 (1/30120), which does not exceed the estimated maximal expected allele frequency for a pathogenic ALMS1 variant of 1/447. A publication cites the variant to have been found in a cohort of Alstrom syndrome patients, however, co-occurrence and cosegregation data was not provided. The variant of interest has not been reported by clinical diagnostic laboratories or databases, to our knowledge. This variant was identified in two individuals with Alstrom syndrome; however, additional details were not provided (Marshall JD et al. Hum. Mutat., 2007 Nov;28:1114-23). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
