The leucine at codon 424 is replaced by phenylalanine, an amino acid with highly similar properties. A different amino acid substitution at this codon (p.L424V) has been shown to segregate with disease in numerous colorectal cancer and polyposis families and to have a deleterious impact on the exonuclease function of the POLE protein (Spier I et al.
