The p.H115P variant (also known as c.344A>C), located in coding exon 2 of the VHL gene, results from an A to C substitution at nucleotide position 344. The histidine at codon 115 is replaced by proline, an amino acid with similar properties. This alteration has been seen in multiple patients meeting diagnostic criteria for Von Hippel-Lindau syndrome who presented with retinal angioma, hemangioblastomas, and renal and pancreatic cysts (Ong KR et al. Hum. Mutat., 2007 Feb;28:143-9; Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic.
