Hereditary cancer-predisposing syndrome;CN230736:Cardiovascular phenotype Uncertain:1
The c.3722G>A (p.R1241Q) alteration is located in exon 28 (coding exon 28) of the NF1 gene. This alteration results from a G to A substitution at nucleotide position 3722, causing the arginine (R) at amino acid position 1241 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.R1241Q variant (also known as c.3722G>A), located in coding exon 28 of the NF1 gene, results from a G to A substitution at nucleotide position 3722. The arginine at codon 1241 is replaced by glutamine, an amino acid with highly similar properties. This alteration has been reported in one proband from a cohort of 279 patients with clinically diagnosed NF1 (Pasmant E et al, Eur. J. Hum. Genet. 2015 May; 23(5):596-601). This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.004% (greater than 110000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence for this variant is conflicting at this time, the clinical significance of p.R1241Q remains unclear. -
