The p.Lys274Thr variant in BSND has not been previously reported in individuals with hearing loss or Bartter syndrome, but has been identified in 1/15294 Africa n chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadins titute.org/; dbSNP rs547338387). Although this variant has been seen in the gene ral population, its frequency is not high enough to rule out a pathogenic role. Computational prediction tools and conservation analysis do not provide strong s upport for or against an impact to the protein. In summary, the clinical signifi cance of the p.Lys274Thr variant is uncertain. ACMG/AMP Criteria applied: PM2 (R ichards 2015). -
