Genes affected
COL11A1 (HGNC:2186): (collagen type XI alpha 1 chain) This gene encodes one of the two alpha chains of type XI collagen, a minor fibrillar collagen. Type XI collagen is a heterotrimer but the third alpha chain is a post-translationally modified alpha 1 type II chain. Mutations in this gene are associated with type II Stickler syndrome and with Marshall syndrome. A single-nucleotide polymorphism in this gene is also associated with susceptibility to lumbar disc herniation. Multiple transcript variants have been identified for this gene. ACMG classification

Classification was made for transcript

Our verdict: Benign. The variant received -13 ACMG points.

BP4
Computational evidence support a benign effect (BayesDel_noAF=-0.91).
BP6
Variant 1-102984046-A-T is Benign according to our data. Variant chr1-102984046-A-T is described in ClinVar as Conflicting_classifications_of_pathogenicity. ClinVar VariationId is 258444.
BS1
Variant frequency is greater than expected in population nfe. GnomAd4 allele frequency = 0.00213 (324/152154) while in subpopulation NFE AF = 0.003 (204/67980). AF 95% confidence interval is 0.00266. There are 0 homozygotes in GnomAd4. There are 171 alleles in the male GnomAd4 subpopulation. Median coverage is 32. This position passed quality control check.
BS2
High Homozygotes in GnomAdExome4 at 5 AR,AD gene
