The p.D685G variant (also known as c.2054A>G), located in coding exon 18 of the MRE11A gene, results from an A to G substitution at nucleotide position 2054. The aspartic acid at codon 685 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Ataxia-telangiectasia-like disorder Uncertain:1
This sequence change replaces aspartic acid with glycine at codon 685 of the MRE11 protein (p.Asp685Gly). The aspartic acid residue is moderately conserved and there is a moderate physicochemical difference between aspartic acid and glycine. This variant is present in population databases (rs554399982, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with MRE11-related conditions. ClinVar contains an entry for this variant (Variation ID: 142045). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
MRE11-related disorder Uncertain:1
The MRE11 c.2054A>G variant is predicted to result in the amino acid substitution p.Asp685Gly. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.024% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-94163093-T-C) and is interpreted as uncertain in ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/variation/142045). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
