This frequency is not significantly higher than estimated for a pathogenic variant in SYNE1 causing SYNE1-Related Disorders, allowing no conclusion about variant significance. c.5462A>G has been reported in the literature in at least one homozygous individual affected with spastic paraplegia and ataxia with other clinical features who carried at least one other variant related to the phenotype in the homozygous state (e.g. Inan_2023).
