MUTYH (HGNC:7527): (mutY DNA glycosylase) This gene encodes a DNA glycosylase involved in oxidative DNA damage repair. The enzyme excises adenine bases from the DNA backbone at sites where adenine is inappropriately paired with guanine, cytosine, or 8-oxo-7,8-dihydroguanine, a major oxidatively damaged DNA lesion. The protein is localized to the nucleus and mitochondria. This gene product is thought to play a role in signaling apoptosis by the introduction of single-strand breaks following oxidative damage. Mutations in this gene result in heritable predisposition to colorectal cancer, termed MUTYH-associated polyposis (MAP). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.971C>T has been reported in the literature in individuals affected with Breast Cancer without strong evidence for causality (Maxwell_2015, Dorling_2021). These reports do not provide unequivocal conclusions about association of the variant with MUTYH-Associated Polyposis or Hereditary Breast and Ovarian Cancer Syndrome. Co-occurrence of the variant with another pathogenic variant has been reported in our laboratory (BRCA1 c.68_69delAG, p.E23fs*17), providing supporting evidence for a benign role.
