The variant allele was found at a frequency of 1.6e-05 in 251204 control chromosomes, found exclusively within the African American population in the gnomAD database. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.10087A>G has been reported in the literature in individuals affected with breast cancer (e.g. Fackenthal_2012, Pal_2015). It has also been reported in 1/2559 African American individuals in the FLOSSIES database (a cohort of women over 70 years of age who have never had cancer). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
