Genes affected
PHOX2B (HGNC:9143): (paired like homeobox 2B) The DNA-associated protein encoded by this gene is a member of the paired family of homeobox proteins localized to the nucleus. The protein functions as a transcription factor involved in the development of several major noradrenergic neuron populations and the determination of neurotransmitter phenotype. The gene product is linked to enhancement of second messenger-mediated activation of the dopamine beta-hydroylase, c-fos promoters and several enhancers, including cyclic amp-response element and serum-response element. Expansion of a 20 amino acid polyalanine tract in this protein by 5-13 aa has been associated with congenital central hypoventilation syndrome. [provided by RefSeq, Jul 2016]
PHOX2B-AS1 (HGNC:40457): (PHOX2B antisense RNA 1)

Uncertain significance, criteria provided, single submitterclinical testingGeneDxFeb 23, 2023In silico analysis supports that this missense variant does not alter protein structure/function; Not observed among a cohort of individuals with multiple primary cancers, but was seen in the unaffected controls (Pritchard et al., 2018); This variant is associated with the following publications: (PMID: 29641532) -
Haddad syndrome Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingInvitaeOct 15, 2023This sequence change replaces threonine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 49 of the PHOX2B protein (p.Thr49Asn). This variant is present in population databases (rs559227588, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with PHOX2B-related conditions. ClinVar contains an entry for this variant (Variation ID: 467719). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt PHOX2B protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Benign:1
Likely benign, criteria provided, single submitterclinical testingAmbry GeneticsAug 18, 2023This alteration is classified as likely benign based on a combination of the following: seen in unaffected individuals, population frequency, intact protein function, lack of segregation with disease, co-occurrence, RNA analysis, in silico models, amino acid conservation, lack of disease association in case-control studies, and/or the mechanism of disease or impacted region is inconsistent with a known cause of pathogenicity. -
