The p.M178T variant (also known as c.533T>C), located in coding exon 5 of the PDLIM3 gene, results from a T to C substitution at nucleotide position 533. The methionine at codon 178 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

The methionine residue is highly conserved and there is a moderate physicochemical difference between methionine and threonine. This variant is present in population databases (rs566270712, ExAC 0.08%), and has an allele count higher than expected for a pathogenic variant (PMID: 28166811).
