The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. The variant was not identified in Cosmic or LOVD 3.0. The variant was identified in control databases in 20 of 276800 chromosomes at a frequency of 0.00007 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: Other in 2 of 6456 chromosomes (freq: 0.0003), Latino in 3 of 34386 chromosomes (freq: 0.00009), European in 9 of 126496 chromosomes (freq: 0.00007), and South Asian in 6 of 30688 chromosomes (freq: 0.0002), while the variant was not observed in the African, Ashkenazi Jewish, East Asian, or Finnish populations. The p.Leu203= variant is not expected to have clinical significance because it does not result in a change of amino acid and is not located in a known consensus splice site. In addition, 5 of 5 in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer, HumanSpliceFinder) do not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time although we would lean towards a more benign role for this variant.
