This changes the amino acid from a glutamine to a stop codon within coding exon 2. In a cohort of 70 Australian breast and ovarian cancer patients, this alteration was identified in one individual with ductal breast cancer diagnosed at age 34 who also had a family history of breast, prostate, and other cancers (Wong MW et al. Breast Cancer Res. Treat. 2011 Jun;127:853-9). Of note, this alteration is also known as 196C>T in published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
