Available data are insufficient to determine the clinical significance of the variant at this time. The frequency of this variant in the general population is uninformative in assessment of its pathogenicity (http://gnomad.broadinstitute.org). Computational tools predict that this variant is damaging. -
Reported previously as a variant of uncertain significance in patients with suspected Charcot-Marie-Tooth disease; however, no additional clinical or segregation information was provided (PMID: 32376792); In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; This variant is associated with the following publications: (PMID: 32376792, Rahman2021[CaseReport]) -

The p.E985G variant (also known as c.2954A>G), located in coding exon 12 of the SH3TC2 gene, results from an A to G substitution at nucleotide position 2954. The glutamic acid at codon 985 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
