This missense change has been observed in individual(s) with cancer susceptible disorder mosaic variegated aneuploidy (MVA) (PMID: 20516114). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs577591919, gnomAD 0.2%), including at least one homozygous and/or hemizygous individual. This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 727 of the BUB1B protein (p.Arg727Cys). -
