KRT1 (HGNC:6412): (keratin 1) The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 482 of the KRT1 protein (p.Tyr482Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of epidermolytic ichthyosis (PMID: 7512983, 22930352). This variant is also known as 481Tyr/Cys. ClinVar contains an entry for this variant (Variation ID: 15909).
