Using alternate nomenclature, this variant would be defined as BRCA2 8296G>A. This variant has not, to our knowledge, been published in the literature as pathogenic or benign. BRCA2 Val2690Ile was not observed at a significant allele frequency in large population cohorts (Lek 2016). Since Valine and Isoleucine share similar properties, this is considered a conservative amino acid substitution. BRCA2 Val2690Ile is located within the DNA binding and the nuclear exporting signal (Yang 2002, Jeyasekharan 2013). In-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. Based on currently available evidence, it is unclear whether BRCA2 Val2690Ile is pathogenic or benign. We consider it to be a variant of uncertain significance. - |
