Studies of a related gene in rat suggest that the encoded protein functions to link clathrin to the sodium channel protein type 10 subunit alpha protein. The encoded protein has also been identified as a component of distal appendages of centrioles that is necessary for ciliogenesis. Alternative splicing results in multiple transcript variants. Studies have shown that this variant is associated with altered splicing, but the impact on the resulting protein product is unknown (PMID: 24285566). Disruption of this splice site has been observed in individual(s) with clinical features of orofaciodigital syndrome or Bardet-Biedl syndrome (PMID: 24285566, 27894351; Invitae). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 5 of the SCLT1 gene. It is expected to disrupt RNA splicing.
