The c.2236_2241delATCATA variant (also known as p.I746_I747del) is located in coding exon 14 of the MSH2 gene. This variant results from an in-frame ATCATA deletion at nucleotide positions 2236 to 2241. This results in the in-frame deletion of an isoleucine at codon 746. This alteration demonstrated decreased protein stability (Ollila S et al. Gastroenterology, 2006 Nov;131:1408-17; Ollila S et al. Hum Mutat, 2008 Nov;29:1355-63). This variant has been identified in multiple individuals who met Amsterdam I/II criteria for Lynch syndrome (Ambry internal data; Ollila S et al. Gastroenterology, 2006 Nov;131:1408-17; Nilbert M et al. Fam Cancer, 2009 Jun;8:75-83). This amino acid region is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the supporting evidence, this alteration is interpreted as a disease-causing mutation. -
