Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251372 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8113G>A has been observed in individual(s) affected with breast and/or ovarian, colorectal, and pancreatic cancers (example: Tung_2015, Tavera-Tapia_2017, Yurgelun_2017, Yu_2022), without strong evidence of causality. These report(s) do not provide unequivocal conclusions about association of the variant with Breast Cancer. At least one publication reports experimental evidence evaluating an impact on protein function.
