This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251372 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8113G>A has been observed in individual(s) affected with breast and/or ovarian, colorectal, and pancreatic cancers (example: Tung_2015, Tavera-Tapia_2017, Yurgelun_2017, Yu_2022), without strong evidence of causality. These report(s) do not provide unequivocal conclusions about association of the variant with Breast Cancer. At least one publication reports experimental evidence evaluating an impact on protein function. Specifically, a pKAP1 phospho-flow assay, calibrated using benign and pathogenic controls, was used to assess whether a variant was functionally neutral or deleterious and this assay demonstrated that the variant of interest was functionally neutral.
