The p.L337F variant (also known as c.1009C>T), located in coding exon 10 of the FANCC gene, results from a C to T substitution at nucleotide position 1009. The leucine at codon 337 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, phenylalanine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
