Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. Disruption of this splice site has been observed in individual(s) with breast cancer (PMID: 30630526). This variant is not present in population databases (ExAC no frequency). This sequence change affects a donor splice site in intron 8 of the FANCC gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. - |

RNA studies have demonstrated that a different alteration impacting the same splice donor site, c.843+5G>A, results in abnormal splicing in the set of samples tested (Ambry internal data). A different alteration at this nucleotide, c.843+1G>A, alteration was identified as a germline finding in 1/66 Chinese triple negative breast cancer patients who underwent tumor and germline next generation sequencing using a 43-gene panel (Yi D et al. Hum. Genomics, 2019 01;13:4). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. - |
