The p.G481C variant (also known as c.1441G>T), located in coding exon 9 of the BRIP1 gene, results from a G to T substitution at nucleotide position 1441. The glycine at codon 481 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in cohort of 1824 patients with triple negative breast cancer who were unselected for family history of breast or ovarian cancer and in 1/854 patients with apparently sporadic pancreatic ductal adenocarcinoma (Couch FJ et al. J. Clin. Oncol. 2015 Feb;33:304-11; Shindo K et al. J. Clin. Oncol. 2017 Oct;35:3382-3390). This variant was also identified in a cohort of 3,579 African males diagnosed with prostate cancer who underwent multi-gene panel testing of 19 DNA repair and cancer predisposition genes(Matejcic M et al. JCO Precis Oncol 2020 Jan;4:32-43). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces glycine with cysteine at codon 481 of the BRIP1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 25452441) and in an individual affected with pancreatic cancer (PMID: 28767289). In an international breast cancer case-control meta-analysis, this variant was detected in 2/60466 cases and 1/53461 unaffected controls (PMID: 33471991). This variant has been identified in 4/243620 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
