This variant has been identified in 4/243620 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The glycine at codon 481 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in cohort of 1824 patients with triple negative breast cancer who were unselected for family history of breast or ovarian cancer and in 1/854 patients with apparently sporadic pancreatic ductal adenocarcinoma (Couch FJ et al. J. Clin. Oncol. 2015 Feb;33:304-11; Shindo K et al. J. Clin. Oncol. 2017 Oct;35:3382-3390). This variant was also identified in a cohort of 3,579 African males diagnosed with prostate cancer who underwent multi-gene panel testing of 19 DNA repair and cancer predisposition genes(Matejcic M et al. JCO Precis Oncol 2020 Jan;4:32-43). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
