The p.I489M variant (also known as c.1467T>G) is located in coding exon 9 of the BRIP1 gene. This alteration results from a T to G substitution at nucleotide position 1467. The isoleucine at codon 489 is replaced by methionine, an amino acid with highly similar properties. Based on protein sequence alignment, this amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces isoleucine with methionine at codon 489 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with tubo-ovarian cancer or breast cancer (PMID: 33471991, 35263119). This variant has been identified in 3/241248 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 241248 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1467T>G has been reported in the literature in at-least one individual affected with Ovarian Cancer (example: Delahunty_2022). This report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Six clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as uncertain significance.
