Breast-ovarian cancer, familial, susceptibility to, 2 Pathogenic:2
Variant allele predicted to encode a truncated non-functional protein. -
The frameshift variant has not been reported previously in heterozygous state in individuals with breast or ovarian cancer Singh J, et al., 2018. The variant is novel not in any individuals in gnomAD Exomes and in 1000 Genomes. This variant has been reported to the ClinVar database as Pathogenic multiple submissios. This variant causes a frameshift starting with codon Serine 163, changes this amino acid to Valine residue, and creates a premature Stop codon at position 9 of the new reading frame, denoted p.Ser163ValfsTer9. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing Borg A, et al., 2010. For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:2
The c.486delG pathogenic mutation, located in coding exon 5 of the BRCA2 gene, results from a deletion of one nucleotide at nucleotide position 486, causing a translational frameshift with a predicted alternate stop codon. Since frameshifts are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
This variant deletes 1 nucleotide in exon 6 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. This variant has been reported in individuals affected with breast cancer (PMID: 25186627, 29470806). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -

Hereditary breast ovarian cancer syndrome Pathogenic:1
For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with breast or ovarian cancer (PMID: 25186627, 29470806). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ser163Valfs*9) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). ClinVar contains an entry for this variant (Variation ID: 254476). -
