This variant is present in population databases (rs587781559, ExAC 0.001%). This sequence change replaces leucine with serine at codon 594 of the BRIP1 protein (p.Leu594Ser). The leucine residue is highly conserved and there is a large physicochemical difference between leucine and serine. -

The leucine at codon 594 is replaced by serine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. To date, this alteration has been detected with an allele frequency of approximately 0.005% (greater than 40000 alleles tested) in our clinical cohort (includes this individual). Based on protein sequence alignment, this amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -
