This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 248320 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.6784G>C has been reported in the literature in at least one individual affected with Ataxia-Telangiectasia, however without strong evidence for causality (e.g., Verlinsky_2007). These report(s) do not provide unequivocal conclusions about association of the variant with Ataxia-Telangiectasia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
