ATM (HGNC:795): (ATM serine/threonine kinase) The protein encoded by this gene belongs to the PI3/PI4-kinase family. This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder. [provided by RefSeq, Aug 2010]

Variant summary: The ATM c.2522A>C (p.Asp841Ala) variant involves the alteration of a conserved nucleotide. 2/4 in silico tools predict a benign outcome for this variant. This variant is not located in any known domain (InterPro). This variant was found in 31/121120 control chromosomes (1 homozygote), predominantly observed in the South Asian subpopulation at a frequency of 0.0018845 (31/16450). This frequency is about 1.9 times the estimated maximal expected allele frequency of a pathogenic ATM variant (0.0010005), suggesting this is possibly a benign polymorphism found primarily in the populations of South Asian origin. This variant has also been reported in two patients with breast or ovarian cancer without strong evidence for pathogenicity (i.e. co-segregation). In addition, multiple clinical diagnostic laboratories have classified this variant as uncertain significance. Taken together, this variant is currently classified as Variant of Uncertain Significance Possibly Benign. -
