This variant has been reported in individuals affected with breast cancer (PMID: 35264596, 38060977). This variant has been identified in 2/245160 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The p.P12T variant (also known as c.34C>A), located in coding exon 1 of the MSH6 gene, results from a C to A substitution at nucleotide position 34. The proline at codon 12 is replaced by threonine, an amino acid with highly similar properties. This alteration was detected in a cohort of 1663 Brazilian breast cancer patients who underwent hereditary multigene panel testing (Guindalini RSC et al. Sci Rep, 2022 Mar;12:4190). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
