The p.L312F variant (also known as c.936A>C), located in coding exon 7 of the ATM gene, results from an A to C substitution at nucleotide position 936. The leucine at codon 312 is replaced by phenylalanine, an amino acid with highly similar properties. This alteration was reported in a study of 1297 cases of early-onset breast cancer and 1121 controls (Young EL et al. J. Med. Genet., 2016 06;53:366-76). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
