Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4.1e-06 in 246166 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1172A>G has been reported in the literature in an individual affected with ovarian cancer (Kraus_2015). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. A ClinVar submission from a clinical diagnostic laboratory (evaluation after 2014) cites the variant as uncertain significance. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 27616075). This variant has been identified in 1/251400 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.Q391R variant (also known as c.1172A>G), located in coding exon 12 of the MLH1 gene, results from an A to G substitution at nucleotide position 1172. The glutamine at codon 391 is replaced by arginine, an amino acid with highly similar properties. This alteration was observed in a patient with ovarian cancer diagnosed at age 53 who did not have a family history of cancer; it was classified as a variant of unknown significance (Kraus C et al. Int. J. Cancer 2017 Jan;140(1):95-102). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
