The variant was absent in 249356 control chromosomes. c.3188C>T has been reported in the literature in multiple individuals affected with Wilson Disease (Loudianos_1999, Weiss_2010, AbdelGhaffar_2011). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 21682854, 10544227, 20517649). Four submitters have cited clinical-significance assessments for this variant to ClinVar after 2014 and classified as pathogenic/likely pathogenic (n=2) and VUS (n=2).
