This missense change has been observed in individual(s) with arrhythmogenic right ventricular cardiomyopathy and/or autosomal dominant dilated cardiomyopathy (DCM) with woolly hair, keratoderma and tooth agenesis (PMID: 22795705, 22949226, 33460606, 34766015). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 564 of the DSP protein (p.Thr564Ile). -
