Rett syndrome Pathogenic:4
This variant has been collected from RettBASE and curated to current modified ACMG/AMP criteria.Based on the classification scheme defined by the ClinGen Rett/Angelman-like Expert Panel for Rett/AS-like Disorders Specifications to the ACMG/AMP Variant Interpretation Guidelines VCEP 2.0, this variant is classified as pathogenic. At least the following criteria are met: Predicted to result in loss of function, and LOF is a known mechanism of disease (PVS1). Has been observed in in at least 2 individuals with phenotypes consistent with MECP2-related disease (PS4_Supporting, PMID: 12707946, RettBase internal database). This variant has been identified as a de novo occurrence in an individual with Rett syndrome without confirmation of paternity and maternity (PM6, PMID: 12707946). This variant is absent from gnomAD (PM2_Supporting). -

not provided Pathogenic:1
Nonsense variant in the C-terminus predicted to result in protein truncation, as the last 34 amino acids are lost, and other loss-of-function variants have been reported downstream in HGMD; Not observed at significant frequency in large population cohorts (gnomAD); This variant is associated with the following publications: (PMID: 18728070, 17684768, 18842453, 11738860, 15691364, 20728410, 18562141, 12707946, 30536762) -
Intellectual disability Pathogenic:1
The variant c.1357C>T, p.(Arg453*) was identified in an individual with neurodevelopmental disorder (NDD) and classified as Pathogenic according to ACMG guidelines. Inheritance for this variant was not maternal. The variant likely explains the NDD in this individual. -
